脾多肽注射液通过肿瘤转移相关蛋白 1 参与大鼠急性肺损伤的保护作用

目的 将脾多肽应用于脂多糖诱导的大鼠急性肺损伤模型,探讨其肺保护机制,为临床应用提供基础研究依据。 方法 将 80 只健康 SD 大鼠随机分为脂多糖组(LPS 组),对照组,脾多肽组和脂多糖+脾多肽组,每组各 20 只。脂多糖(LPS)5 mg/kg 通过腹腔内注射的方式建立大鼠急性肺损伤模型。6 h 后麻醉处死大鼠获取肺组织,通过 ELISA、湿干重比值、HE 染色、TUNEL 以及 Western-blot,检测肺损伤相关指标。 结果 脾多肽显著改善肺组织损伤,减轻肺水肿,抑制炎症因子的释放和细胞的凋亡,此外,脾多肽可抑制肿瘤转移相关蛋白 1(MTA1)的表达。 结论 脾多肽注射液对大鼠急性肺损伤具有保护作用。

Objective To analyzed the role of lienal polypeptide injection in acute lung injury induced by lipopolysaccharide (LPS) in rat. Methods Eighty male SD rats were randomly allocated into 4 groups: a LPS group, control group, lienal polypeptide group and LPS+ lienal polypeptide group (20 rats in each group). Lienal polypeptide or normal saline was given with an intramuscular injection 30 min after an intraperitoneal injection of LPS (5 mg/kg). The severity of pulmonary injury was evaluated 6 h after LPS challenge. Results Lienal polypeptide injection treatment significantly attenuated LPS-induced pulmonary histopathologic changes, alveolar hemorrhage, and neutrophil infiltration. Moreover, the lung wet-to-dry weight ratios, as the index of pulmonary edema, were markedly decreased by lienal polypeptide injection. In addition, lienal polypeptide injection decreased the productions of the proinflammatory cytokines including tumor necrosis factor alpha, interleukin 1β and apoptosis. Furthermore, lienal polypeptide injection significantly suppressed LPS-induced activation of metastasis associated protein-1. Conclusion Lienal polypeptide injection has been demonstrated to protect rats from LPS-induced ALI by the expression of Metastasis-associated protein 1 (MTA1).
